UDP-glucose: flavonoid 3-O-glucosyltransferase (UFGT) is a key enzyme in anthocyanins biosynthesis pathway, which catalyzes the transfer of the glucosyl moiety from UDP-glucose to the 3-hydroxyl group of anthocyanidins, producing the first stable anthocyanins. To elucidate the effect of UDP-glucose: Flavonoid 3-O-glucosyltransferase (UFGT) gene on anthocyanin accumulation in Prunus persica fruit peel, this study isolated a UFGT gene from peach fruit peel by using RT-PCR techniques, which named as PpUFGT. Results show that the full-length cDNA of PpUFGT was 1866 bp containing a 1455 bp open reading frame (ORF) encoding 485 amino acids with a calculated molecular mass of 54.34 kDa and an isoelectric point of 6.03. The protein predication of PpUFGT showed that there were a function domains GT1_Gtf_Like and typical structure of Glycosyltransferase_GTB_type superfamily. Predicted protein secondary structure showed that PpUFGT protein contents of the structure with alpha helix (Hh) 46.49%, extended strand (Ee) 15.10%, random coil (Cc) 24.38%, and beta turn (Tt) 9.50%. PpUFGT shared high similarities to angiosperm UFGT and displayed the closest genetic relationship to Prunus mume. These results indicate that PpUFGT belongs to UFGT families and responsible for anthocyanin accumulation in peach fruit peel.
